I generally use PaintTool Sai. For some reason Fire Alpaca doesn't work properly for me.

Gimp. I haven't tried many though but it seems to do everything I need and it's free. Need to support more free and open source software.

Seems all of you use SAI. :3 I think is one of the best programms around the net,Photoshop CS5 for effects and things like that. I have also FireAlpaca, even thought I don't use it so often, but is a mix of SAI and Photoshop if I can say it,the stroke is good, and the lines are not so 'Hard' as SAI. GIMP...Well,I used it in the past as I said...But it crash always, especially if you draw in a big canvas D: (My size of canvas is always 3500x3500 meh.)

Currently I just use Gimp. Before that I was using Sketchbook mobile on my phone to do digital art. So much pinch zooming
